Background and Aim: Abstract Breast cancer is one of the most common malignancies that has serious effects on women´s health. The woman's lifetime risk for developing breast cancer is about ۱۰%. The UCA۱ gene produces a long non-coding RNA that have been diagnosed in various cancers, such as breast cancer. Food-derived flavonoid quercetin, has therapeutic potential in cancer, cardiovascular and neurological diseases. The aim of this study was to evaluate the effect of quercetin on the expression of LncRNA UCA۱ in breast cancer cells.Methods: The effect of quercetin on the expression of UCA۱ gene in MCF-۷ cells of breast cancer was assessed by treating cells with different concentrations of quercetin at ۲۴h via MTT assay, IC۵۰ was determined; RNA was extracted using triazole and then cDNA synthesized. The expression levels of UCA۱ gene relative to GAPDH gene at different concentrations of quercetin were quantified using the highly sensitive Real Time- PCR method.Results: The results showed that quercetin has dose-dependent inhibitory effect on the viability of MCF-۷ cells. The quantitative real-time polymerase chain reaction results showed that UCA۱ gene expression was significantly (p=۰.۰۰۱) downregulated in the presence of quercetin.Conclusion: Quantitative real-time PCR results showed that Qurcetin could decrease MCF-۷ cell number by decreasing UCA۱ gene expression.
