Up-regulation of hsa-miR-۵۵۸۴-۵p in breast cancer and association with lymph node metastasis and higher grades

Background and Aim: Breast cancer is the second leading cause of cancer deaths in women. miRNAs are small non-encoding molecules that regulates cellular processes and their expression is dysregulated in human cancer through various mechanisms. miR-۵۵۸۴ has been associated with an increased risk in some disease, However its function and expression in breast cancer has not been studied yet. The aim of our study is to evaluate miR-۵۵۸۴ expression in breast cancer and association with clincopathological factors.Methods: In this case-control study, ۳۵ FFPE tumoral tissues of breast cancer and ۳۵ non-tumoral adjacent tissues were enrolled. Followed were informed consent and completing clinical information for all samples. Total RNA was extracted and specific complementary DNA (cDNA) for miRNA was synthesized, afterwards the relative gene expression for miR-۵۵۸۴-۵p was determined using qRT-PCR and evaluated by ۲^(-∆∆ct) method. Finally, the expression pattern was analyzed by statistical analysis.Results: The results indicated that, the average of miR-۵۵۸۴ relative expression was significantly higher in tumor tissues compared to adjacent non-tumoral and there was statistically a significant difference between them with a ۹۵% confidence level (P=۰.۰۰۱). The results also indicate a significant increase in expression of lymph node metastasis tissues (P = ۰.۰۳). In this study, the up-regulation of miR-۵۵۸۴ was observed in tumor grade ۲ and ۳(P = ۰.۰۱).Conclusion: The results of recent study have revealed miR-۵۵۸۴-۵p may function as an oncogene in breast cancer. Therefore elevation of the expression miRNA is associated with increased cell proliferation and invasion. Overexpression of miR-۵۵۸۴-۵p could contribute to the pathogenesis of breast cancer.
