چکیده مقاله:
Background and Aim: Gene therapy is a new branch of medical sciences that is a new and effective approach to the specific treatment of human genetic disorders. Gene therapy can be substituted for classic chemotherapy as a potentially successful method. Non-viral nanocarriers today with a wide variety of compounds and biological properties are widely used for gene delivery in vitro and in vivo. Liposomes are one of the transmission systems that have received much attention as potential carriers for a wide range of therapeutic agents. Liposomes are capable of delivering hydrophilic and lipophilic therapeutic agents due to the dual properties of their constituents.Methods: In the present study, to synthesize monolamellar liposomes, a lipid film hydration method was used that, during hydration, a pCAG-GFP-puro expression vector containing the GFP gene was inserted into the liposome. These plasmid-containing monolamellar liposomes were transferred to MCF-۷ breast cancer cell lines and finally, their toxicity was evaluated in cancer cells.Results: The results indicated that these nanocarriers have the potential to transduce the pCAG-GFP-puro vector and have no toxicity effect on the cells, therefore, it can be further investigated as a safe gene delivery system.Conclusion: The results indicated that these nanocarriers have the potential to transduce the pCAG-GFP-puro vector and have no toxicity effect on the cells, therefore, it can be further investigated as a safe gene delivery system.
