PAX۸ gene expression analysis in thyroid neoplasm

Background and Aim: Thyroid neoplasm is the most common endocrine cancer in the world and its growing incidence has become a worldwide issue. PAX۸ gene regulates thyroid specific genes and also genes with important roles in the thyroid follicular cells function. The aim of this study is to evaluate the potential of PAX۸ gene for the diagnosis of the thyroid neoplasm.Methods: Thyroid neoplastic and their neighboring healthy tissues were collected from Sina hospital, Isfahan, Iran. RNA extraction, cDNA synthesis and RT-qPCR reaction were performed using specific exon-junctioned primers. Beacon V۸.۲۱ software and Oligo V۷.۰ software were used for specific primers designing and confirmation. REST ۲۰۰۹ software would be used for data analysis.Results: PAX۸ gene (NCBI-ID: ۷۸۴۹) analysis by STRING database revealed the potential role of the selected gene in thyroid homeostasis. KEGG databases also showed that PAX۸ could cause thyroid cancer development. The designed primers had specific binding only to PAX۸ gene,appropriate ۳’?G and they lacked secondary structures such as hairpins. Agarose gel electrophoresis (۱%) showed the acceptable quality of the extracted RNA. The A۲۶۰/A۲۸۰ ratio was ۱.۸-۲.۲ that indicate the purity of the extracted RNA.Conclusion: PAX۸ gene as an important regulator of vital functions such as transcriptional activity, cell proliferation and apoptosis, plays important roles during thyroid tumorigenesis. Some studies have been shown that PAX۸ expression was decreased in follicular carcinoma. In addition the PAX۸–PPARγ fusion oncogene is common in follicular thyroid cancer but not in other type of thyroid neoplasms. It must be mentioned that the pattern of PAX۸ expression is still unclear. Therefore in this study we would investigate the amount of PAX۸ expression in cancerous and neighboring healthy tissues to determine the role of PAX۸ in thyroid tumorigenesis and cancer development.
