A bioinformatics analysis of exosomal microRNA released by breast carcinoma: Looking for a diagnostic biomarker

Background and Aim: Breast cancer is the most common cancer among women worldwide and one of the leading causes of cancer deaths. The origin of breast cancer is breast tissue, so studying breast tissue is of specific importance for the prognosis, diagnosis and treatment of breast cancer. Exosomes have recently been recognized as an important player in breast cancer pathogenesis. They are secreted nanoparticles that are present in both normal and pathophysiological conditions in all body fluids and are also effective in cell-cell communication. microRNAs (miRNAs, miRs), as the most important cargo in exosomes, are potential cancer biomarkers that can be useful in diagnosis and prognosis. To finding a potent diagnostic biomarker, we checked the EVmiRNA, a database of miRNA profiling in extracellular vesicles and re-analyzed the expression profiles of specific exosomal miRNAs in breast cancer that were present in three samples.Methods: The expression of exomiRs was studied using the EVmiRNA database. This database provided ۴۶۲ small RNA sequencing samples of EVs from ۱۷ sources/diseases and constructed the EVmiRNA database (http://bioinfo.life.hust.edu.cn/EVmiRNA) to show the miRNA expression profiles. We analyzed SRR expression levels in breast carcinoma in EVmiRNA database (SRA id: SRR۱۹۷۳۹۵۷ and SRR۱۹۷۳۹۶۲). We also checked the expression of our candidate miRNAs in ExoCarta database (http://www.exocarta.org/).Results: Our bioinformatics analysis revealed that miR-۳۱۶۸ was overexpressed in exosomes derived from breast carcinoma samples compared to normal exosomes. The expression of exomiR-۳۱۶۸ was also found in low levels in normal plasma according to the Exocarta database.Conclusion: Given the high expression levels of miR-۳۱۶۸ in breast carcinoma exosomes, our study suggests that miR-۳۱۶۸ could serve as a potent and non-invasive diagnostic biomarker for breast carcinoma.
