Antileishmanial activity of Carum copticum extract against Leishmania (L) tropica [MHOM/IR/92/SHIRAZ] promastigotes: An Ex vivo study

Subject & aims: Leishmaniasis is one of the ten important infectious diseases of the world which is common in our country in the three widespread spectrum of cutaneous, visceral, and Diffused Cutaneous Leishmaniasis disorders. Glucantime is the selective drug for Leishmaniasis with limited administration due to its toxicity and untoward complications and also its resistance against some parasites. This study investigated the Antileishmanial activity of Carum copticum extract against Leishmania (L) tropica [MHOM/IR/92/SHIRAZ] promastigotes: An Ex vivo study.Materials & Methods: The standard species of Leishmania tropica was transferred to RPMI1640 culture medium. Then, the effect of different concentrations of Carum copticum extract on Leishmania promastigotes was compared to Glucantime in the static and dynamic stages using the MTT colorimetric method with ELISA. Next, the survival percentage of the parasite was statistically analyzed.Results: The Carum copticum extract and Glucantime were challenged with urban Leishmaniasis parasite at 24, 48, and 72 h which were repeated three times. The 4.68 μg/L concentration was the minimal concentration of Carum copticum extract that exerted the greatest significant effect on the parasite in the static phase at 24 h. The 250 μg/L concentration of Glucantime was the minimal significant concentration that was effective at 48 h in the static phase of the parasite (P<0.05).Conclusion: The Carum copticum extract exerts an effect on the survival of urban Leishmaniasis promastigotes and stops their growth. Hence, future studies should focus on wider exploration of the effect of this extract on its in vivo survival.
