This study was carried out to find the optimum environmental conditions for propagation of Chiku on Kerole rootstocks through in vitro micrografting technique. Uncoated Kerole seeds and shoot tips (4.4% contamination) and single nodes of Chiku were successfully surface sterilized in 10% commercial bleach for 16 min. A hundred percent seed germination was obtained in media containing 3 levels of sucrose in presence of 1/2, 1/3 and full MS salts strength. The highest epicotyl and root length, and the highest number of lateral roots were produced when seeds were germinated on medium containing 15 gL-1 sucrose at 1/2,1/3 and 1/2and 1/3 strength of MS salts, respectively. No auxiliary shoots were initiated from shoot cultures on MS medium containing BA at 0.0 to 2.5 mgL-1, IBA or NAA, Kinetin 0.0 to 1.5 mgL-1 in media containing 30, 40 and 50 gL-1 sucrose. In such media a clumps of callus was formed at the basal ends of shoot explants and began to split the surrounding tissues. Due to failure in shoot proliferation required as scion donor for micrograftiong, this study did not progress further.
