Green synthesis of silver nanoparticles using methanolic extract of Berberis vulgaris L. fruit and its antimicrobial activity

Iran Background and Objective: Microbial resistance against antibiotics is growing recently. There have been new reviews of silver nanoparticles that have been synthesized by the mediation of plant extracts. These silver nanoparticles can be considered as an alternative treatment of various microbial infections. The objective of this study is to find out the antimicrobial effect of Berberis vulgaris L. silver nanoparticles. Materials and Methods: The synthesis of silver nanoparticles in this study has been done by using a methanolic extract of Berberis vulgaris fruit, which was prepared by maceration technique In this study, the best condition of the green synthesis of nanoparticles was established. The particles morphology and size have been investigated by micrographs. Moreover, the antimicrobial effect of the particles have been studied using minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) assays. Five gram-negative bacteria strains such as Acinetobacter baumannii (ATCC 19606) and two gram-positive bacteria strains such as Staphylococcus aureus (ATCC 29213) were used in the study. Findings: The optimum condition of this green synthesis was room temperature, best pH 10, optimum concentration of silver nitrate was 10 mM and 30 minutes was the best time for the synthesis. These nanoparticles made by green synthesis from methanolic extract of Berberis vulgaris leaves showed the highest antimicrobial activity for Pseudomonas aeruginosa (MIC= 0.39μg/mL, MBC= 3.12μg/mL) in compared to Ciprofloxacin (MIC=512 μg/mL). Ciprofloxacin was used as a positive control in the MIC and MBC test. The average size of nanoparticles was 28.6 nm showed by the SEM test. Conclusion: According to the results of this study, silver nanoparticles made from methanolic extract of Berberis vulgaris fruit synthesizes by green synthesis method showed very good antimicrobial activity and can be an alternative product for antibiotics.
