Relationship between GPR54 gene rs3827421 polymorphism and incidence of infertility in women of West Azarbaijan province

Background and Aim : Couples failing to conceive after one year of regular intercourse withoutpreventing infertility say the GPR54 gene is located on the long arm of the human chromosome 2, whichhas 5 exons and 5 introns and has a length of 1179Kb and a protein of 368 amino acids. A protein thatcontains two membrane-bound fragments and three glycosylation sites. Mutation in this gene causes earlycentral maturation (CPB).Methods : In this study, 95 infertile women from Urmia city were divided into control group and 110healthy blood samples were divided into patient group. DNA extraction was performed by salt depositionmethod for PCR amplification and sequencing. Sequences of RFLP technique were used and finally datawere analyzed using SPSS V.24 software.Results : The mean age of the patients was 52 7 7 years. Frequency of C allele was 69% and T allele was31% which was significantly correlated with its frequency (P = 0.05). Also heterozygous (TC) genotypeswere 51% (P = 0.03), while dominant homozygotes (CC) and recessive homozygotes (TT) were 19% and30%, respectively, indicating no association with disease incidence. Also (P> 0.05), in women who had afamily marriage 2% and women who had a family history of the cancer had a 1% incidence compared toother individuals and groups (P = 0.04) (CL = 1.19-3.71- OR = 95%).Conclusion : Based on the results of the rs3827421 polymorphism mutation of GPR54 gene can be usedas one of the preliminary identification factors.
