FTIR Spectroscopic Pattern of Royan Mouse Embryonic Stem Cells at various stages of differentiation

Introduction: Understanding of the different steps in Mouse Embryonic Stem cells differentiation is one of the most complicated and yet unclear parts of the biology with a great impact on Teratology in the toxicology science. The aim of this study was to introduce FTIR as a new technique for the recognition of the progress and development of stem cells differentiation.Methods: The mouse Embryonic Stem Cells have been acquired from Royan Research Institute and differentiated into beating heart cells. FTIR spectroscopy in the range of 3600-600 cm-1 has been performed on these samples for 16 days. 30 spectra were taken on each day and averaged spectra for each day were calculated. These spectra were baseline corrected, differentiated and the peaks picked carefully.Results: There were some shifts in bands 1013cm-1, 1612cm-1, 1674cm-1, 2910cm-1 & 2965cm-1. Those are related to produce of nucleotides to Z form and then Z to A form DNA, as well as the extensive formation of line and sheet form proteins during the differentiation that are the important findings using FTIR spectroscopy.Conclusion: This investigation may present the potential of this technique for the identification of the embryonic differentiation step, as well as the molecular and cellular alterations of the embryonic cells during this period. This might indicate of great applications in biology particularly embryology and teratology.
