Effects of three retrograde filling materials on production of inflammatory cytokines

Background and Objective: Root-end resection surgical procedures include applying a root-end filling material to seal efficiently, and to enable the periradicular tissues to repair and regenerate. These materials should not trigger inflammation processes. Pro-inflammatory cytokines such as IL-1β and TNF-α are playing crucial roles in host anti-microbial and anti-tumor response, but production of these cytokines during inflammatory processes can stimulate bone resorption. In this study, the effects of the three types of root-end filling agents on pro-inflammatory cytokine production was investigated by human normal monocytes and macrophage cell line (THP-1) after 24 hours’exposure.Matherials and Methods: In the present study, THP-1 monocyte cell lines beside peripheral blood monocytes (PBM) were used. Peripheral blood mononuclear cells (PBMCs) were isolated by Ficoll gradient centrifugation of buffy coat. The experimental materials after mixing and setting were exposed to UV rays and then to the prepared cells. After 24 hours supernatants were collected and kept frozen at -20oc. Cytokine concentrations were measured by ELISA KitsFindings: The exposure of monocytes to EBA for 24 hours caused decrease in TNF-α and IL-1β secretion compared with other groups. In THP-1 culture with materials, release of pro-inflammatory cytokines was not significantly different between groups. Our results indicated that Geristore caused the lowest TNF-α production, and in secretion of IL-1β, it was after EBA.Conclusion: Taken together, our results indicated that Geristore had almost poorer performance in production of inflammatory cytokines.
