Introduction and Aim Breast cancer is one the most common malignancy among women worldwide. One of the major problems in cancer therapy is the incomplete eradication of the invasive primary tumor mass or dissemination of tumor stem cells, leading to recurrence of disease. Targeted therapy of cancer using oncolytic viruses has generated much interest over the past few years in the light of limited efficacy and side effects of standard cancer therapeutics for advanced diseases. The purpose of this study was to evaluate the efficacy of HSV-G47delta oncolytic virus (3rd generation of engineered HSV-1) for the treatment of breast stem cells.Methodology Stem cells isolated from MDA-MB-435 cell lines using sefum free media supplemented with EGF,FGF, LIF and b27. Stem cells identified using two specific surface markers including, CD44 and CD24.Inoculation of g47delta in stem cells performed with 1.5MOI of concentration and gene expression analysis took placed in a standard real time protocol.ResultsGene expression analysis of oncolytic virus effects, revealed that g47delta potentially targets breast cancer stem-like cells using Ras protein and the expression of Ras protein in this signal was significantly chabged Conclusion The inhibition of cancer growth and progression is one of the major challenges faced by modern medicine. HSV-G47delta able to kill breast cancer stem cells by targeting Ras signaling pathway. Our preliminary data supports continuing the development of CSC targeting and safe treatment of breast cancer.
