It’s been proved that polymorphism in PRNP gene is the most principal factor and determinative in scarpie disease resistance that three loci (154,136,171) have the main function. In this study was conducted PRNP polymorphism on 49 Zell breed sheep. DNA extraction was carried out by salty optimized method in animal science biotechnology laboratory University of Tehran. Genotyping of the animals was done with PCR-RFLP method for three loci related to scrapie disease. Results of Genotyping for the locus of 136 showed only AA genotype for all of the animals which were monomorph in this locus and also all of the animals were monomorph in locus of 154 with RR genotype. But three genotypes were observed in locus of 171(H, Q, R). The genotype of ARQ/ARQ had the most genotype frequency (0.408) and other genotypes frequencies of ARR/ARH, ARQ/ARH, ARQ/ARR, ARQ/ARQ and ARH/ARH are 0.102, 0.102, 0.326, 0.408 and 0.061 respectively. According to investigations it’s been proved that genotypes ARR/ARH, ARR/ARQ are resistant to scrapie and genotypes ARH/ARH, ARH/ARQ don’t have significant difference in comparison with other two groups (sensitive to scrapie and insensitive to scrapie). Genotype of ARQ/ARQ which observed in Zell sheep is sensitive to scrapie. Due to importance of the genotype and because of its high frequency in population and relationship with scrapie sensitiveness, must be considered in selection programs.
