Chitosan / Poly (L-lactide) polymeric nanofibers containing Imipenem antibiotic: synthesis, antibacterial properties and drug release for wound dressing

Synthesis of biocompatible polymer nanofibers is valuable, due to their use as a cover for burns and as a replacement for bandage because of their antimicrobial properties. In this study, electrospinning of chitosan and nanofibers synthesis with antibacterial properties was investigated. Nanofibers with antibacterial properties were synthesized by electrospun of chitosan/poly (L-lactide)/imipenem polymer solution. The results showed that the optimized ratio of chitosan/ poly (L-lactide) polymer solution was ratio of 50 to 50 and chitosan 2 wt.-% and poly (L-lactide) 10 wt.-% polymer solution was the best weight percentage for nanofiber preparation with the mentioned aims. Also, the average diameter of Ch/Pla/Imi nanofibers was 143 nm and measured with ImageJ software. Afterwards, the antibacterial properties of imipenem as additives (with different percentages) was studied in the polymer solution. The scanning electron microscopy (SEM) images and antibacterial tests were showed that the electrospun of Chitosan/Poly (L-lactide)/Imipenem polymeric nanofibers were effective against gram negative bacteria Eshrishia coli and gram positive bacteria Staphylococus aureus and inhibited growth of gram negative bacteria E. coli. After that, placing Ch/Pla/Imi nanofibers in to Phosphate buffer pH=5.5, measuring the absorbance of drug release in phosphate buffer, showed that release of drug from nanofibers had been continued until one weak. Thus, it can be a good candidate for bandages because of long release of antibacterial drug to prevent bacterial infection in wounds.
