The Effect of Olive Leaf Extract on Vascular Endothelial Growth Factor Gene Expression in Ischemic Muscle of Hind Limb Ischemic

Introduction: In normal conditions, body can improve ischemic tissues by creating collateral vessels (angiogenesis). Vascular endothelial growth Factor (VEGF) is one of the most important factors affecting angiogenesis. The olive leaf extract have antioxidant and anti-inflammatory effects. In this study, the effect of olive leaf extract on VEGF gene expression was investigated.Methods: Twenty four male wistar rats were randomly divided into four groups. Sham (surgery without femoral artery ligation + normal saline), Sham + olive leaf extract (300 mg/kg/day, gavage for 21 days), Hind limb ischemia (surgery with femoral artery ligation + normal saline), Hind limb ischemia+ olive leaf extract (300 mg/kg/day, gavage for 21 days). After 21 days the rats were sacrified and gastrocnemius muscle removed to evaluate the VEGF gene expression by Real Time PCR method.Result: In this study gene expression of VEGF in the ischemic muscles of treatment group (300 mg/kg olive leaf extract) was significantly lower than the sham group.Conclusion: In this study, it seems that olive leaf extract reduced the VEGF gene expression. Therefore, it is possible that the olive leaf extract exerts its beneficial effects on ischemic tissues other than increasing VEGF.
