Molecular Identification of the Wolbachia spp. in the Culicidae Genera collected from Firozabad of Fars, Iran 2019

Background: The three main culicidae genera, Culex , Anopheles and Aedes transmit lymphatic filariasis, malaria and dengue fever respectively. Recent culicidae control strategies have demonstrated unsuccessful, and there still is a considerable number of morbidity and mortality from mentioned diseases. Genetic control rules have now appeared as new plan of actions. Fluorobacterial identification of mosquitoes and awareness of the presence of Wolbachia spp can help to genetically modify bacteria in the control of vector-borne diseases by paratransgenic method, symbiotic bacteria are genetically modified and reestablished in vectors, where they express effective genes.Objectives: This study was carried out to detection of Wolbachia spp. in Culicidae Genera collected from Firozabad of Fars.Materials and Methods: Totally 8 and 5 genomic DNA were pools (including 10-12 mosquitoes for Culex and Anopheles species respectively). The mosquitoes were catched and identified from rural regions of Firozabad, were investigated based on surface protein gene (wsp) by Polymerase chain reaction (PCR) method. Four primers were designed by Oligo0.7 and GeneRunner 0.4 software on the wsp gene including 2 forward and 2 reverses as outer and inner primers. The Nested PCR was done to confirm the positive samples by inner primers. The expected size bond in this study were 650 and 430 bp with outer and inner primers respectively.Results: Out of 4 of the 8 culex and 1 of the 5 of Anopheles examined genomic DNA pools were found to be positive, 50.0% and 20% respectively. The obtained sequences were alignment by NCBI online site and show that have similarity to other Wolbachia strains available in the GenBank. According to the phylogenetic tree comparing the sequences showed similarity with Group B wolbachia. The present research reveals the first molecular detection of Wolbachia endobacteria in Cx. laticinctus populations in Iran.Conclusion: Due to the scientists approach to new methods and to use the paratransgenic techniques, it is necessary to identify the bacterial flora of Iranian vectors for the control of vector borne diseases
