Molecular and Structural Features of Netrin-1 in Lucilia sericata Larvae as a Potential for Wound Healing

Background: Today, L. sericata larvae are used for maggot therapy. The Netrins are from the laminin family which are secreted from the L. sericata larvae salivary glands. Netrin-1 has the most important role in the Axonic guidance And it s an enzyme that causes Neuroregeneration, angiogenesis and migration cell, also play an important role in wound healing and biomarker in diagnosis of some diseases. Objectives: The aim of this study was to identify molecular and structural feature of Netrin-1 in L. sericata larvae. Materials and Methods: L. sericata larvae were reared in standard condition. Then by methods of Rapid Amplification of cDNA Ends (RACE) and Rapid Amplification of Genomic Ends (RAGE), nucleic acid sequence of L. sericata Netrin-1 were identified. Parts of the gene including, the middle part, 3′ end, and 5′ end, were identified and TA cloned in PTG19 then transformed to DH5ɑ Ecoli, each part sequenced, then assembling was done by SeqMan software. Results: The Full length of mRNA sequence of Netrin-1was 2451 bp. In addition, ORF was identified with 2115 bp by Oligo0.7 software that can be translate Netrin-1 protein with 704 amino acid residues. Molecular weight was estimated 78.6 kDa. Three-dimensional structure of Netrin was drawn by SWISS-MODEL and were showed that Netrin-1of L. sericata has similar to Netrin-1 of human, with 61.8% coverage. Conclusions: Netrin-1 protein can help repair the myelin membrane in the neuronal cells. Ultimately it can be a good candidate for Multiple Sclerosis treatment and wound healing. Also our next step is to produce recombinant proteins for use in medical sciences
