Background and Objectives: Gaillonia aucheri Jaub. &Spach (Rubiacea), is a woody shrub growing in south of Iran. The aim of the present work was to evaluate the toxicity of this species to normal and cancerous cell lines. Methods: The aerial parts were collected from Bandar abbas, Hormozgan province. The plant parts were coarsely powdered and 10 g of the dried powder was extracted with methanol 80 % by maceration method. The cytotoxic activity of the extract was evaluated through MTT method which is a colorimetric assay that shows the cell metabolic activity. The viable cells are able to convert MTT to formazan crystals; thus, the results of this method can be interpreted about cytotoxicity of a material to the exposed cells. The effect of the extract was evaluated against MDBK (Bovine kidney cells), HT-29 (colon carcinoma), MCF-7 (human breast adenocarcinoma), HepG-2 (human hepatocellular carcinoma) and A-549 (non-small cell lung carcinoma) cell lines. Results: Gaillonia aucheri extract exhibited no cytotoxic activity to any of the above mentioned cell lines up to the concentration of 300 μg/mL. Conclusion: Considering the results of the present study, G aucheri was not toxic to the cell lines regarding the tested concentrations.
