Up-regulation of lncRNA PVT1 correlates with advanced stages and invasion in ESCC patients.

Introduction & Aim: Esophageal Cancer (EC) as a lethal malignancy with lower survival rate is considered as an incurable disease. Abnormal expression of long non-coding RNAs in cellular processes were reported in EC. lncRNA PVT1 (Plasmacytoma Variant Translocation 1) locates on 8q24 region of human chromosome which is known as an oncogenic lncRNA. Association of lncRNA PVT1 with cancer development and pathophysiology was demonstrated. This investigation aims to explore the expression level of PVT1 in ESCC tissue. Methods: The expression level of PVT1 was in measured ESCC and margin non-tumor tissues (n=40), through the extraction of total cellular RNA, synthesis of cDNA and RT-qPCR method. Meanwhile the correlation between this gene expression with clinical parameters in ESCC patient was analyzed. Results: PVT1 was significantly up-regulated in ESCC tissues compared with non-tumor tissues. Up-regulation of PVT1 was positively associated with advanced stages (T3 and T4) and distance metastasis. (p<0.05). Conclusion: PVT1 was up-regulation in ESCC patients and is associated with advanced stages and distance metastasis. These results may indicate the potential role of PVT1 in ESCC and its possible regulatory role. Thus, further studies are required to identify the possible oncogenic role of this lncRNA and its regulatory mechanism. Exploration of molecular mechanisms behind PVT1 may be useful in designing diagnostic bio-markers and therapeutic interventions.
