چکیده مقاله:

IntroductionLittle progress have been achieved recently in Glioblastoma (GBM), 4TH grade glioma, treatments. Dysregulation of several microRNAs expression, small noncoding RNAs, in GBM have been reported before. Overexpression of some microRNAs lead to more proliferation, invasion and metastatic behavior in GBM and Inhibition of these oncomiRs in cancer researches resulted in remarkable results. Exosomes, 30-100 nm extracellular vesicles, attracts lots of attention today and have the potential to carry different cell components. Exosomal transfer of microRNAs in tumor microenvironment was observed, too.ObjectivesHere we attempt to suppress upregulated oncomiRs in GBM cells by using exosomal transfer of anti-miRs.MethodsAnti-miRs transfected to HEK-293T and after producing stable cell lines, exosomes was extracted. GBM cell line (U87) treated with extracted exosomes then and microRNA expression level was analyzes.ResultsOur result revealed the exosomal transfer of anti-miRs construct can downregulate microRNA expression and overexpress miR target genes.ConclusionThese findings guide us toward new therapeutic agents for gene therapy, exosomes, for cancer treatments with any kind of side effects because of using natural cell products and carriers
