IntroductionApplying stem cell to adjust injured tissue has been used in recent years. Therefore, the tissue engineering field shows different potentiality for therapeutics and regeneration in neurological disorders. AlsoAdipose-derived stem cells (ADSCs) are an easily available source of Mesenchymal stem cells. ADSCs can transdifferentiate into cells of other lineages such as neural stem cells.MethodsIn this study, ADSCs were isolated and cultured from rat adipose tissue by digestion with collagenase, ADSCs at passage 3 were identified by immunocytochemistry with antibodies analysis against CD44, CD105, as well as CD45, CD106. We could induce rat ADSCs into floating neurospheresand neurospheres evaluated by immunocytochemistry for NF68 and Nestin.Diameter and number of this Neurospheres evaluated. Neurosphere can be converted into neural stem cell, NSCs were confirmed by immunocytochemistry with NF68, nestin.ResultsThe isolated ADSCs expressed CD44 and CD105, but they proved negative for CD45, CD106. Neurospheres evaluated by immunocytochemistry that expressed NF68 and Nestin. We observed that the diameter of neurospheres increased and the number of Neurosphere decreased. Immunocytochemical assessment of NSCs showed positive for Nestin and NF68.ConclusionThe present study demonstrated that ADSCs can be efficiently induced into neurospheres under appropriate conditions. Moreover, these neurospheres can then be expanded and converted into neural stem cells (NSCs).
