Fabrication and characterization of nanobiocomposites based on Iota-carrageenan/silk fibroin nanofibers for bone tissue engineering

Bone tissue has one of the highest demands for tissue reconstruction and bioresorbablescaffolds because of high rate of bone fracture and high prevalence of bone defects. Biodegradable natural polymers are developing to fabricate scaffolds. Furthermore, the combination of protein-polysaccharide in scaffolding together with the ability to induce bone-like apatite formation has become a promising approach to mimic extracellular matrix composition. In this study we developed and characterized new bioactive composite scaffold from iota-carrageenan (i-CG)/ (SF) nanofibers for bone regeneration application. Different ratios of fibroin nanofibers (5, 10 mg) were added to 1ml of i-CG solution to enhance mechanical and biological properties. Then 3D scaffolds were fabricated via freeze-drying technique. Various characterization techniques were applied to analyze such items as the structure, morphology, compressive strength and cell toxicity assessment. An interconnected porous structure suitable for cell penetration was proven by scanning electron microscopy. Compressive tests were carried out in static mode under dry condition that revealed a strong impact of scaffold because of good mechanical properties of fibroin nanofibers. Also osteoblast-like cell (MG-63) culture experiments indicated that all scaffolds are biocompatible.
