Studying the suppressing effect of mesenchyme stem cells derived from amniotic membrane (AMMSC) on colorectal cancer (HT-29 cell line)

Introduction Colorectal cancer is known as the third deadly cancer in the world. Nowadays, there are many approaches to treat cancer. Mesenchyme stem cell (MSCs) therapy is considered as a proper tool for biological activities and treatment of some non-malignant and malignant diseases. Due to the unknown effect of the mesenchyme stem cell on cancer, especially colorectal cancer, this study examines the inhibition effect of AMMSC on colorectal cancer (cell line HT29). Method First, MSC was isolated from the human amniotic membrane and then, performed identity tests. The isolated AMMSC were co-culture with HT29 cell line and then MTT assay was used to determine the proper inhibitory concentration of MSC on HT29 cell line. Immunocytochemistry assay was performed to measure the expression of CDK2 protein, the Scratch test was used to evaluate the effect of the MSC on the HT29 cell migration and for evaluating gene expression of bcl2 in apoptosis and p21 in the cell cycle tumor cells were used realtime-PCR assay.Result Co-culture was performed by MTT test, a proper concentration of 1:5 fold for 72h was determined for the inhibition effect of MSC on tumor cells. Immunocytochemistry test results indicated the protein expression of CDK2 in tumor cells was reduced, The Scratch test proved that MSCs significantly inhibited the HT29 cell migration. Real time-PCR results showed the quantitative gene expression of Bcl2 was reduced and increased gene expression of p21 in tumor cells .Conclusion The result showed that the AMMSCs play an important role in suppressing colorectal cancer cell line HT29 progression by inhibiting angiogenesis, suppressing cell cycle and Induction of apoptosis, Therefore AMMSC is a proper cell for cell therapy
