The effect of nanofiber policaprolacton scaffold on the chondrogenesis of infrapatellar adipose tissue derived stem cells in restoration articular defects in vivo

IntroductionIn the present study, we report the development of a nanofiber polycaprolactone scaffold that can function as a stem cell carrier to induce chondrogenesis and promote cartilage repair in vivo.MethodsInfrapatellar fat pads were obtained from sheep knee and the stem cells were isolated and characterized by flow cytometry then at a concentration of 1 ×106/100μl were seeded into polycaprolactone. four defects were created in sheep knee, two defects received adipose tissue derived stem cells-polycaprolactone construct, second group received the polycaprolactone (PCL), the third group was chosen as a ASCs group and other defects as a fourth group was kept empty in during the study.ResultsMorphological evaluation showed that defects treated with ASCs-scaffold constructs were completely filled with cartilage-like tissue, while in ASCs group, PCL group and empty defects revealed the formation of a thin layer of cartilage-like tissue in the defects. Analyze of Real-Time RT-PCR showed the increase in collagen type 2 mRNA levels, aggrecan and Sox9 in ASCs/PCL groups to compared with the other groups.Immunofluorescence results pointed to protein expression of collagen type 2 in ASCS/PCL groups, while in PCL groups, ASCs group and empty defect group were not appeared type II collagen protein and aggrecan.Toluidine blue staining showed the formation of round and polygonal clusters of chondrocytes in ASCs/PCL construct groups and fibrocartilage tissue in other groupsConclusionApplying the stem cell–scaffold combination caused the articular cartilage defect repair that appeared in our results. Therefore, we showed that nanofiber polycaprolactone promote the chondrogenesis of infrapatellar adipose tissue derived stem cells in vivo
