Simultaneous Determination of Aflatoxins in Pistachio Nuts Employing Gold-labeled Antibody Biosensor and Multivariate Calibration

This report is aimed at comparison of the results obtained during spectrofluorimetric and HPLC analyses of aflatoxins B1, B2, G1 and G2 in pistachio nuts. Application of antibody coated gold nanoparticles (GNPs) specific to aflatoxins B1, B2, G1 and G2 in rapid aflatoxins determination method was presented in this paper. Due to severe overlapping between the emission profiles for the aflatoxins, they cannot be determined by direct inspection of data. The strategy used in this study, constituted by partial least square (PLS) regression, provided to be easy to implement and to originate reliable results. After sample treatment and without any extended cleanup steps and derivatization process, PLS is applied on full-spectrum. PLS can be successfully applied on spectrofluorimetric data for simultaneous determination of totalaflatoxins. HPLC analyses were carried out using a symmetry C18 column and a mobile phase composed of methanol– water (40:60, v/v), with a flow rate of 1.0 ml/min and fluorescence detector. The outcome of spectrofluorimetric and HPLC measurements were well correlated. There are no significant differences between their results. Proposed method may be a good alternative to the traditional methods of aflatoxins analysis
