Efficacy and Safety of PRP Therapy for Patients with Knee Osteoarthritis

Background and Aim: Platelet-rich plasma (PRP) has been frequentlyused to regenerate tissue damages in different organs such as skin,mucosal membranes, and joints. In this study, we sought to investigatethe regenerative capacity of PRP to treat cartilage lesions of patients withknee osteoarthritis (OA).Methods: In this phase 1 clinical trial, 14 patients with mild to moderateknee OA referred to the Rheumatology Clinic in Imam Reza Hospitalwere enrolled. All patients received a single intra-articular injection of 2mL of autologous PRP. All subjects were visited serially 1, 3 and 6 monthsafter treatment and were evaluated for the amount of pain (assessed withvisual analog scale), joint stiffness, the range of motion and crepitus.Results: The PRP treatment was found to be safe and no adverse eventsoccurred. Significant and almost linear improvements in stiffness andrange of motion were observed. In addition, pain and crepitus decreasedsignificantly during the 6-month follow-up. Multivariate analyses showedno significant impact of age, gender, and BMI on the level of clinicalresponse.Conclusion: The intra-articular injection of PRP showed clinicalimprovement in patients with mild to moderate knee OA and producedno adverse events. This provides the evidence for efficacy and safety ofPRP as an alternative regenerative treatment for knee OA.
