Background and Aim: Human Mesenchymal stem cells (hMSCs) are ofgreat interest due to their high potency to differentiate. The behavior ofthese cells depends on the paracrine and biophysical signals received fromthe 3D microenvironment. The simultaneous use of suitable nanofibrousscaffolds and conditional media can simulate the microenvironmentneeded by stem cells. The aim of this study is to fabricate and evaluatethe effect of the Tissue-engineered scaffold of Poly (L-lactic acid) (PLLA)and conditional medium on mesenchymal stem cell culture.Methods: Poly (L-lactic acid) (PLLA) nanofibrous porous scaffolds wereprepared using by electrospinning method. The structure of the scaffoldswas investigated using scanning electron microscopy. The identity of bonemarrow-derived hMSCs was confirmed by osteogenic and chondrogenicdifferentiation and flow cytometric analysis and then cultured in the PLLAscaffold in the presence of MSCs conditional medium. Cellular adhesionof hMSCs was assessed using by SEM. The viability and proliferation ofthe cells were then determined by an MTT assay.Results: SEM data confirmed the porosity of scaffold with suitable poresize. Histological and flow cytometric analysis confirmed bone marrowderivedhMSCs. The MTT assays revealed the biocompatibility of PLLAnano scaffold under conditional medium.Conclusion: The 3D architecture of PLLA nanofibrous scaffolds cansupport the viability and proliferation of hMSCs under conditionedmedium.
