Study of the Association Between Polymorphisms of Nrf2 and HIF1a Genes with the Growth Rate Of Umbilical Mesenchymal StemCells

Background and Aim: Mesenchymal stem cells have the ability todifferentiate into different cell types and have immunosuppressiveproperties and the ability to migrate and replace in the affected area. The Nrf2 and HIF1a genes are the main regulators of oxidative stress andinflammation. The aim of this study was to determine the associationbetween the polymorphisms of Nrf2 and HIF1a genes with the growthrate of umbilical mesenchymal stem cells.Methods: In this study, we used three recombinant plasmids, includingpcdna3.1-Nrf2 and three recombinant plasmid pcdna3.1-HIF-1α for thetransfection. The extracted mesenchymal cells from the umbilical cordwere transfected for the expression of Nrf2 and HIF1a genes in cells thatwere genetically modified and tested by real-time PCR. The cells werethen examined using the WST-1 method and cell survival after exposureto stress was investigated in terms of against oxidative stress and oxygendeficit and serum poverty.Results: The survival rate of umbilical mesenchymal stem cells with Nof2and HIF1a genes (89.3% ± 2.9%) were more than the control group (49.7± 28%). Also, the cells with Nosf2 and HIF1a genes encountered thestresses (6.9 ± 21%) were more than the control group (51.9 ± 6.2%). Theexpression of Nrf2 and HIF1a genes in comparison with the oxidativegroups with the oxidative stress groups was markedly higher in the S-HIFNFR2gene than other groups (82% ± 7.7%). The survival rates of NSCHIFgenes were the lowest as compared to the other genes (65.2% ±6.7%) in the MSCHIF-NRF2 oxygen deficiency group. The genes had ahigh survival (86.5% ± 8.9%) and in serum (P> 0.05). The frequency andsurvival of the genes were higher than the other genes (28.7 ± 7.9%).Conclusion: Based on the results, the survival rate of the umbilicalmesenchymal stem cells with the Nrf2 genes and HIF1a was increasedagainst oxidative stress and oxygen deficit and serum poverty.
