Methods: Thirty-five rats were randomized into 5 groups (n = 7): 1) Defect alone (control), 2) PVA/ VBP scaffolds (NP-), 3) PVA/ VBP /nGO scaffolds(NP+), 4) PVA/ VBP scaffolds with hASCs cultured 7 days in osteogenicmedia (NP-osteoblast-like cells), and 5) PVA/ VBP /nGO scaffolds withhASCs cultured 7 days in osteogenic media (NP+ osteoblast-like cells).An 8 mm critical-size circular calvarial defect was made in each rat. Thescaffolds belong to each group implanted in the defects and follow up for6 to 12 weeks. The animals were sacrificed and bone repair was evaluatedusing multiple computerized tomography (CT) and hematoxylin-eosin(H&E) staining at 6 and12 weeks in all experimental groups.Results: These data provide evidence that constructs made of NP+ andNP- along with osteoblast-like cells transplantation have great boneregeneration capability in critical-sized calvarial defects than constructswithout osteoblast-like cells. NP+ osteoblast-like cells-treated animalsshowed significantly improved regeneration of new bone formationcompared to those in the other groups (P<0.05). Histological analysisshowed that cells migration and healing process in rat calvarial defecttreated by NP+ osteoblast-like cells and NP- osteoblast-like cells werehigher than in the other groups.Conclusion: PVA/ VBP /nGO scaffolds with osteoblast-like cells mayhave wide applicability for bone repair as an alternative bone graftmaterial alone.
