Background and Aim: Human amniotic membrane (hAM) is theinnermost layer of fetal membranes, which surrounds the developingfetus and forms the amniotic cavity. hAM and hAM-derived cellspossess unique properties that make them excellent candidates for usein regenerative medicine, such as low immunogenicity, promotion ofepithelization, anti-inflammatory and antimicrobial properties as well asangio-modulatory, anti-fibrosis and anticancer properties. We examinedthe effect of human amniotic epithelial cells- conditioned media (hAECs-CM) on cancerous cell lines proliferation.Methods: Human placentas were obtained at term pregnancy duringCaesarean sections from women with negative for HIV-I, and hepatitis Band C. The amnion was manually separated from the chorion and washedextensively in PBS. The epithelial cells were isolated enzymatically.Briefly, amnion was treated with 0.15% trypsin-EDTA for 15 and 40 min at 37°C. Tissues were then filtered through a 70μm cell strainer to collectsingle cell suspension and centrifuged at 1200 rpm for 10 min. The cellpellet was resuspended and cultured in flask 25 in complete culturemedia. The medium was changed once after the density of cells reached85% confluency, and hAEC-CM was collected 48 h later while cellswere growing exponentially. The hAEC-CM were then collected, 0.22mm filtered, and used in subsequent experiments. Four different cancercell were seeded in 96-well tissue culture plates. After 24h the culturemedium was exchanged with hAEC-CM. After 48h of the incubation, cellviability was determined by the MTT assay.Results: Percentage cell viability of breast cancer cell lines includesMDA-MB-231, MCF-7, and BT-474 recorded by 67%, 73%, and 56%,respectively. Amongst four cell lines, Hela (cervical cancer cells) showedthe least viability by 38%.Conclusion: Our results indicated that hAECs-CM able to inhibit theproliferation of cancer cells. Taken together, these results provide strongevidence that hAECs-CM can be used as a safe and effective cancertargetingcytotherapy for treating breast and cervical cancer.
