Much attention has been given to understand how cellular processes arechanged in the neurological diseases, which have long been hampered bythe inaccessibility of the brain. Nevertheless, some recent technologicaladvancements and progresses appear to be in favor of overcoming thishurdle. Of these, one is the ability to direct the differentiation of patientspecifichuman pluripotent stem cells (hPSCs) into the neuronal and glialcells/tissue. Editing specific loci in the genome using programmablesequence-specific nucleases provides another possibility, which mightallow the precise removal or addition of mutations of interest. In thistalk, I will discuss the progress made in establishing isogenic hPSC-basedmodels of neurological disorders. I will also articulate their utility inunderstanding neuronal dysfunction during development and disease.
