Hepatitis B virus (HBV) infection is the leading cause of hepatocellularcarcinoma (HCC) worldwide. However, due to its species and organspecificity, and limitations in model systems to study the virus, themolecular events that drive HBV-mediated HCC remain unclear. Wehave established and characterized 3D human liver organoids as aprimary platform to model HBV infection-induced tumorigenesis. Liverorganoids are seeded from infected patient liver biopsies and used inthe molecular and functional analysis. We find that patient-derived liverorganoids resemble their primary tissue of origin, and display an earlycancer gene signature, presenting a patient-specific in vitro platform fordisease surveillance and drug discovery.
