Cytotoxicity of Methanol Extract of Cichorium Intybus Aerial Parts on Embryonal Carcinoma P19 Cells

Background: Cichorium intybus is a medicinal plant that has anti proliferative effect on some cancer cell lines. Embryonal Carcinoma (EC) cells are pluripotent cells interatocarcinomas and are generally considered to be archetype of cancer stem cells (CSCs). CSCs are drug resistance cancer cells with ability of self-renewal and differentiation.Therefore, targeting CSCs provides a novel therapeutic strategy for cancer treatment. Objectives: The aim of this study was to investigate ant proliferative activity of methanolextract of aerial parts of C. intybus on EC P19 cell lines as a model of CSCs. Materials/Patients and Methods: P19 cells were cultured in minimum essential mediumeagle, alpha modification with 10% fetal bovine serum methanol extract of aerial parts of C. Intybus were obtained and cytotocic activity of extract was investigated in variousconcentrations (10, 20, 40, 80, 160, 320, 640, 1280 and 2560 µg/ml) after 48, 96 and 144h incubation. MTT assay was carried out to analyze cell viability.Results: The IC50 of methanol extract of aerial part of C.intybus were determined as 1253, 727 and 568 µg/ml after 48 h, 96 and 144h of treatment which indicate weak cytotoxicactivity on P19 cell Conclusion: MTT results showed that methanol extract of C.intybus L aerial parts did not exhibit an acceptable cytotoxic activity on embryonal carciboma cell line . According to another researches, aqueous extract of C.intybus, exhibited a modest inhibition in cell proliferation for tree cancer cell lines prostate, breast and colorectal. CSCs are pluripotent cancer cells with self-renew and differentiation capacity into non-CSC progeny. Moreover, CSCs, have a higher intrinsic resistance to conventional therapies through a variety of mechanisms. This resistance may lead to low cytotoxicity of C.intybus L on P19 cell line
