Background: Brucellosis, an endemic and important zoonotic disease in Iran that must be controlled since it responsible for a considerable economic and health-related burden. Brucellosis primarily affects domesticated livestock and humans can also be infected via the consumption of contaminated meat or dairy products. The rapid and accurate identification of brucellosis is one of the most important measures to control the disease in humans and animals,so this study was intended to investigation of new methods for the identification of Brucella strains at genus and species level. Methods: A systematic review of twenty databases was searched, with 119 articles published between January 1994 –June 2017 about different methods for identification of brucellosis and information was analyzed by SPSS software. Results: In general, there are two methods to identify the strains of Brucella, in the first category, using classical typing methods included sensitivity to dyes and production of H2S, differentiation among Brucella species or biovars is made possible. In the second category the use of molecular techniques includes: AMOS PCR, REP-PCR, RAPD-PCR, RFLP-PCR, Bruce-ladder multiplex PCR, Real-time PCR, MLVA, HOOF-print, HRM analysis, ERIC-PCR, SNP-based MGB PCR and MLST are operate to identify the genus and species of Brucella. Conclusion: Genotyping methods (MLVAand MLST) are useful to trace-back epidemiological analysis and it will be used as routine typing techniques to allow the identification of Brucella strains at genus and species level within a given biovar in future.
