Study of antimicrobial activity of Bacillus megaterium isolated from honey

Background: Nowadays, multi antibiotic-resistant pathogens are an important health problem in medicine. Therefore finding novel sources of antimicrobial compounds is essential to introduce effective agents against resistant pathogen microorganisms. One of the microorganisms producing antimicrobial is Bacillus. The present study looks to antimicrobial activity of Bacillus megaterium, which isolated from honey from south of Iran. Methods: The B. megaterium was cultured into a broth medium. After incubation the bacterium was grown for 48h at 30 °C. The bacterial extracts were filtered through a 0.22 µm pore filter membrane after centrifugation of bacterial culture at 5000 rpm for 10 min. Six microorganisms were studied as human pathogenic organisms and 8 were used as plant pathogenic. All bacterial strains were cultured on nutrient agar and fungal strains were cultured on MEA media. The microbes were dissolved in sterile distilled water and compared to 0.5 McFarland control. The plates were then incubated at 30 °C for 24 hours and the diameter of the zone of inhibition was measured. Results: The antimicrobial test showed that B. megaterium has ability to produce antimicrobial against plant pathogens. This isolate showed the highest inhibition zone against Fusarium oxysporum. Discussion: The Discovery of novel antibiotic is essential subject because probability of multiple resistances on pathogenic microorganisms is very high. This is the first report on the antibiotic production ability of B. megaterium.
