Histopathological modifications of the rat prostate following oral silver nanoparticles administration

Background and Objectives: Silver Nanoparticles (AgNPs) have recently received much attention for their possible applications in medicine, cosmetics, textiles, electronics, and other fields. However, little is known about its toxicity in reproductive organs of animal model. This study therefore, tried to examine the effects of silver nanoparticles on the histopathological changes in the prostate of rats. Materials and Methods: In this experimental study, 40 adult male Sprague-Dawley rats were divided into five groups. In the four experimental groups, nanosilver particles were given orally for 28 consecutive days at doses of 30, 125, 300, or 700 mg/kg. Rats in the control group received equal volumes of deionized water. After paraffin embedding and Hematoxylin and Eosin (H&E) and masson’s trichromestaining staining, histological changes evaluated in prostate. Photomicrographs were obtained for morphometric analysis motic camera. Data were analyzed using one-way analysis of variance (ANOVA). Results: The obtained results showed an increase in collagen sedimentation in 30 and125 mg/kg of AgNPs groups (p=00.00). The epithelium height and size of epithelial nuclei were also increased significantly 30 and125 mg/kg of AgNPs groups in compare to control (p=00.00). The other histological changes that evaluate in the prostate of rats affected by AgNPs included: decreased alveolar secretory folded and increased vacuoles in the epithelial cells, thickness of muscle fibers and the ratio of parenchyma to stroma. These histological alterations were also more evident in rats exposed to 125 and 30 mg/kg of AgNPs. Conclusions: Based on the present study, it can be concluded that oral administration of silver nanoparticles for 28 days had effects on prostate that could show the toxicity of Ag NPs.
