Background: Pleomorphic adenoma (PA) is the most common salivary gland tumor. Mucoepidermoid carcinoma(MEC) and Adenoid cystic carcinoma (AdCC) are the most common malignant salivary gland tumor. Cyclooxygenase-2 (COX-2) is a key regulatory enzyme that its overexpression in various tumors is also correlated with progression, proliferation, metastasis and inhibition of apoptosis Vascular endothelial growth factor(VEGF) is a growth factor and it is a potent angiogenic mediator that has a important role in neoplastic angiogenesis.The main aim of this study was combined immunohistochemical analysis of cyclooxygenase-2 with vascular endothelial growth factor expression in Pleomorphic adenoma, Mucoepidermoid carcinoma and Adenoid cystic carcinoma of salivary glands. Material and methods: A total of 90 specimen of 30 PA, 30 MEC and 30 AdCC were collected from department of pathology Amiralam hospital .Immunohistochemical staining was performed using antibody againts COX-2 and VEGF.Chi-square and Gamma analysis were used for data analysis. Results: statistically significant correlation was apparent between frequency and intensity of COX-2 expression and intensity of VEGF expression in PA , MEC and AdCC but not about frequency of VEGF expression.In this study significant correlation was seen between frequency of COX-2 and VEGF in MEC and AdCC but not in intensity of this tumors. Indeed, significant correlation was not seen between frequency and intensity of COX-2 and VEGF with grade of histology and lymph node metastasis of MEC and AdCC. Conclusion: The significant correlation of VEGF expression with COX2 expression may represent the role of COX2 in tumor angiogenesis by modulating VEGF production
