WT1 over expression in FAB different subtype of acute myeloid leukemia and its relationship with other laboratory parameters

Background Several studies have shown the oncogenic properties of WT1 in some tumors. WT1 with anti-apoptotic activity takes part in the expansion of myeloid cells,and its expression is inversely related to cell differentiation.The majority of patients with AML have high levels of WT1 expression. This study was to investigate the expression of WT1 in the different FAB subtype of acute myeloid leukemia (AML) and its relationship with other laboratory parameters. Methods: Ninety de novo patients with AML and 20 healthy donors were enrolled in this study, the WT1 expression in peripheral blood mononuclear cells was detected by TaqMan RT-PCR. The correlation of WT1 with other laboratory parameters of AML patients was analyzed. Levels of WT1 were expressed as copies of WT1/ABL × 104 and a copy number ≥250 was considered asoverexpression. Results: The frequency of WT1 overexpression at diagnosis in the current study was 84%. The WT1 expression level in de novo AML patients was higher than that in healthy donors (P < 0.01). FAB-AML-M3 patients exhibited higher expression of WT1 than that in M1, M2 and M4 (P < 0.05) . No significant difference was found between WT1 expression levels and clinical characteristics except in WBC count (p=0.04). No correlation was found between WT1 expression levels and achieving a first CR after induction therapy (p=0.5). Conclusion: Our findings confirmed that WT1 be a pan marker for monitoring of acute myeloid leukemia patients especially those who have no specific chromosomal translocations or other molecular marker.
