Expression of cancer stem cell marker ALDHl, correlated with high grade prostate cancer

چکیده مقاله:

Introduction:Prostate cancers contain a small population of cancer stem-like cells which have roles incancer initiation and progression.Aldehyde dehydrogenase (ALDH1) activity has been used to isolate stem cells of variouscancer and normal cells. In this study we evaluated the expression pattern and clinicalsignificance of cancer stem Cell (CSC) marker, ALDHl in a series of prostate tissues.Material & Methods:Prostate tissues composed of 110(68%) prostate cancer (PCa), 21(13%) high-grade prostaticintraepithelial neoplasia (HPIN) and 31(19%) samples of benign prostate hyperplasia (BPH)which were included in tissue microarray have been immunohistochemically examined forthe expression of ALDHl. The correlation of ALDHl expression with clinicopathologicalparameters was also assessed.Results:Our study demonstrated that ALDHl localized mainly in the cytoplasmic area of tumors.There was a significant difference in expression of ALDHl in adenocarcinoma tumorscompared to HPIN and BPH (p-value<0.001).The expression of ALDHl was significantlyhigher in adenocarcinoma tumors, moderate in HPIN and weaker in BPH. In addition,ALDHl expression was positively correlated with gleason score (P = 0.015).Conclusions:Our finding showed that expression of ALDHl in prostate cancers is significantly high incomparison to HPIN and BPH and increasing level of Gleason score has a direct correlationwith expression of ALDHl marker that means tumor become more aggressive and tendencyfor metastasis. Therefore this cancer stem cell marker can be a potential marker for targetedtherapy of malignant tumors of prostate.
