COMPARISON OF FLOW CYTOMETRY AND PANEL REACRTVE ANTIBODY METHODS IN DETECTION OF ANTI-HLA-CLASS IANTIBODIES

Objective: To compare Anti-HLA antibodies detected by flowcytometry and Panel ReactiveAntibodies (PRA) test.Samples and Methods: In this study Anti-HLA antibodies in sera of 82 patients suffer fromdifferent hematological disorders (like: ALL, AML, Aplastic Anemia and ITP) and 44subjects (included: 30 candidate subjects for renal transplantation and 14 multiparouswomen) were detected by flowcytometry and PRA.Results: our results showed 44 out of 82 (53.7%) patients showed Anti-HLA class-Iantibodies by flowcytometry and which 15 of these patients (18.3%) had positive PRAresults. All of Controls were positive with 2 methods. Frequency of each antibody isopytewas found the following percent: IgM (51.2 %), IgG (32.9%) and IgA (1.2 %) by flowcytometry. There was a good relationship between 2 methods (P=0.0001, r=0.55).Conclusion: Despite of a good relationship between PRA and flow cytometry methods indetection of Anti-HLA Class I antibodies, PRA can only detect antibodies with Complementfixation characteristics.
