Various isolation procedures of blastomeres and the developmental capacity of blastomeres derived from eight cell mouse embryos to blastocyst stage

چکیده مقاله:
Introduction: The propose of this study was to investigation and compare of developmental potential of isolated blastomeres derived eight cell mouse embryos using various chemical and mechanical methods of separating blastomeres. Methods and materials: In this study, different protocols for isolating individual blastomeres out of eight cell mouse embryos were used. The individual blastomeres were obtained by exposure to tyrode acid, Hyalronidase, EDTA or by mechanical force. Then the blastomeres were cultured in vitro to blastocyst stage. The total number of blastocysts was counted and the diameter of blastocysts was measured. Results: The mean and standard deviation of the total number of cells in the mechanical method, thyroid, EDTA and hyaluronidase solutions were 42.2 ± 3.39 ± 2.39, 42.9 ± 2.85, 39.9 ± 2.85 and 41.8 ± 2.18 respectively . The mean and standard deviation of blastocyst diameter in the above groups were 78.81 ± 5.14, 75.18 ± 14.21, 77.25 ± 5.22, and 22.25 ± 5.9, respectively. Conclusion: All isolation procedures had a negative impact on the in vitro growth patterns of isolated blastomeres but the mechanical group was shown better development to blastocyst stage. Our results were not explained severe retardation of isolated blastomeres derived from eight cell mouse embryos.
