Introduction & Aim: Breast cancer is one of the most common types of cancer among women and the second leading cause of cancer-related deaths in the world. It is frequently treated with chemotherapy and hormone therapy. More recently, however, ‘targeted therapy’ has emerged as an important approach to cancer therapy. Curcumin is a potential agent for both the prevention and treatment of cancers. The main way on which the curcumin causes cancer celles to be inhibited is not specified.The main goal of this study was to analyse gene expression changes after treatment of T47D cells with curcumin to investigate signaling pathways involved in inhibition of breast cancer cell growth.Materials and Methods: To evaluate cytotoxic effects of curcumin in T47D cancer cells, MTT assay was done. And then RNA-Seq used for transcriptom profiling. Results: MTT assay showed that the IC50 of T47D cell line for 48h was 30 µM. Transcriptome analysis by next-generation sequencing (RNA-seq) showed that cell cycle regulated genes are main target of curcumin action.Conclusion: RNA-Seq showed that curcumin is a potent agent in the induction of cell cycle arrest with favorable pharmacological activity,and curcumin may have potential as an effective cancer therapeutic for breast cancer.
