Introduction & Aim: Several studies have been shown the role of radiotherapy in cancer treatment for definitive and palliative strategies in variety types of cancer. The conventional cytotoxic agents are used as a radiosensitizer to increase the cancer cells sensitivity to radiation. However, they increased the sensitization of both normal and tumor cells. To overcome this limitation, many studies have conducted to use agent with specificity for tumor cells or low toxicity. The aim of this study was to evaluate the effect of combined bromelain and irradiation therapy on the in vitro survival of breast cancer cells 4T1.Methods: Mouse breast cancer cell line (4T1) was treated in 6-well plate for 24 h with bromelain (95 µg/ml). Then the cells were exposed to different doses of 0, 2, 4 and 6 Gy of 6 MV photon (Clinical accelerator Siemens, located in Parsian hospital, Shahrekord, Iran). The efficacy of combined treatment was investigated using a clonogenic assay. Kruskall-Wallis and Dunn test were used to analyze statistically differences..Results: The survival of 4T1 cells was decreased after combined treatment in a number and size dependent manner with respect to the control group (P<0.05). Survival fraction of 0.51, 0.17, and 0.03 were resulted for doses of 2, 4 and 6 Gy respectively.Conclusion: The combination of irradiation therapy and bromelain has led to a significant reduction in the number, growth and survival of 4T1 cell. Other cell biology techniques should be used to confirm these results.
