Effect of the hyperthermia on the treatment of MCF-7 breast cancer cell line in the presence of gold nanoparticles

Introduction & aim: Hyperthermia is a treatment technique used to increase the tumor temperature from 37 ° C to 40-45 ° C. In conventional hyperthermia methods, the highest amount of heat reaches to the surface of the target and decreases with increasing distance from the surface, furthermore, these methods do not make any difference between healthy and tumor tissue, causing a lots of side effects. Recently, nanotechnology has been used to overcome these limitationsfor specializingtreatments. The purpose of this study was to investigate the effect of hyperthermia and Gold Nano Particles (GNPs) for the treatment of MCF-7 breast cancer cells based onthe clonogenic assay.Methods: The cells were incubated with 20 nm and 20 mg/LGNPs for 24 hours, and then exposed to RF capacitive hyperthermia with a power of 150 W and 13.56 MHz for 2,5 and 10 minutes. All thetreatmentgroupswere incubated for 14 days. Thereafter, Their surviving fractions were calculated and normalized to the control group.Results:The survival rate in the treatment groups was reduced compared to the control group. Between the treatment groups, the group receiving hyperthermia for 10 minutes, the percentage of viability was less than the others.Conclusion: Our data showed the effectiveness of 20 nm GNPs with a20 mg/L concentration in enhancing the effect of RF hyperthermia carried over for 10 minutes.
