Involvement of Pak1 gene expression in cytotoxicity of verbascoside on MDA-MB-231 breast cancer cells

Abstract: Introduction & Aim: Breast cancer is a current malignancy in woman. Considering Pak1 are involved as metastasis markers through the polymerization of actin filaments, its inhibition may be valuable against tumor invasion. Thus the aim of this investigation is studying the effect of verbascoside as natural compound on Pak1 signaling pathway.Methods: MDA-MB-231 breast cancer cells were cultured in RPMI-1640 medium containing 10% FBS and 1% penicillin and streptomycin at 37 ˚C. After treatment MDA-MB-231 cells with various concentrations of verbascoside, MTT assay was performed to determine cytotoxic effect. Then, scratch test was used to evaluate the cells invasion and Real Time PCR was conducted to measure Pak1 expression in MDA-MB-231 cells.Results: The scratch test indicated that verbascoside dose-dependently reduced MDA-MB-231breast cancer cells survival and migration. The results of Real Time PCR indicated that verbascoside reduced Pak1 mRNA level.Conclusion: verbascoside induced breast cancer cell death and suppressed MDA-MB-231breast cancer cells invasion by involvement of Pak1 as an imperative metastasis and invasion marker.
