Sequences, molecular and protein structure bioinformatics analysis of the NGF gene in goats

Nerve Growth Factor (NGF) is a 26 kDa protein that belongs to the beta nerve growth factor(β NGF) family that is required for the survival, maintenance and development of discrete neuronal populations in the nervous system [1] Also, The major site of synthesis of NGF is the ovary, and NGF is responsible for ovarian innervation, follicular assembly and folliculogenesis in animals such as sheep and goat [2,3] . The aim of the present study was to sequences characterization, molecular bioinformatics analysis of the NGF gene in reproductive of goats. The DNASTAR (DNASTAR Inc., Madison, WI, USA), and Genomics (CLC Genomics, version 4) softwares were used for finding cis-regulatory elements in genomic sequences, CpG islands and ORFs regions. Also, the nucleotide sequences were translated using ExPASy translates tools website. The secondary structure protein (3D) was predicted using the proved self-optimized prediction method (SOPMA). Sequencing analysis showed that the promoter and exon 3 regions of the NGF gene contained of conserved motifs such as several regulatory elements, one CAAT motif (at position 79 to 83), one synonymous (g. G > A) substitution and one non synonymous (g. C > T) substitution which, changes positively charged Arginine to nonpolar Tryptophan) mutations(1) the according to (Gen Bank accession no. KF724722.1). Significant increase in the expression of FSH and LH receptor genes in the oviduct after treatment with NGF also indicated the role of NGF in regulating the reproduction via the interactions with gonadotropins [4] . Secondary protein structures (3D) was predicted for exon of NGF gene that included this amount; βturns (11.20) αhelices (18.67), extended strands or βfolds (28.22) and random coils (41.91), respectively. On the other hand, results indicated that there is one possible CpG islands (CGIs) in the guanine- and cytosine rich regions for epigenetic and genome studies. Maximum likelihood estimate of substitution matrix has shown that nucleotides have an equal united of maxim and minim, respectively. Regarding to the results of present study; it seems that more studies are needed to evaluate the NGF gene with prolificacy.
