چکیده مقاله:
Development of industry and technology leads to biological issues due to presence of pollutants such as aromatic hydrocarbons with mutagenesis and carcinogenic potential. Hence, providing strategies toreduce the resource or degradation of this type of compounds based on biological methods could be a promising strategy for prevention of related diseases hat has been considered in this study. Therefore,the purpose of this study was to investigate the structural and functional properties of fungal lignin peroxidase in order to develop the prebiotics and transformation of probiotic strains capable of eliminating aromatic hydrocarbons based on modern in-silico methods.In this regard, the protein sequence of lignin peroxidase from Phanerochaete chrysosporium was retrieved from NCBI databank with accession number AAD46494.1, and characterized its physicochemical properties. Moreover, homologues proteins of desired enzyme and correlation of them were determined based on BLOSUM62 matrix by Blast and MEGA 6 programs, respectively. 3D structures of CotA and its homologues sequences were achieved by SWISSMODEL server. Subsequently, GROMACS and AutoDuck software were used for assessment the thermo stability and affinity of these enzymes to PAHs.The results of this study can be concluded tertiary structures of homologues sequences of lignin peroxidase enzyme in different fungal strains with suitable quality, which are selected the best of them based on stability at 70 ° C and high-affinity substrate for synthesis were considered for more biotechnological process.
