چکیده مقاله:
Cancer is the result of an imbalance in cell proliferation and death. many factors contribute to the incidence of cancer. One of the most important strategies in cancer therapy is the cell death induction,increased antioxidant capacity of cells, and inhibition of angiogenesis. Therefore, the use of compounds with any of these effects can be useful in the treatment of cancer. Nowadays, in the nano industry,materials are synthesized that have great potential in cancer treatment. Some of these compounds are potent antioxidants, and others can suppress cancer cells with the least side effects on other cells.Therefore, this study was designed to evaluate the antioxidant and cytotoxic properties of nanoparticles.ABTS•+ was prepared by combining ABTS solution (7 Mm) with potassium persulfate (2.4 Mm) 16 hours in the dark. Then the solution was diluted 50 times to produce a work solution. The stock solution of the ZnO NPs were prepared and a serial dilution was performed to prepare various concentrations (62.5-500 μg/ml) of the ZnO NPs. The volumes of nanoparticle and ABTS free radicals were transferred to a microtip, after an incubation time, their absorbance was measured at 734 nm. For MTT test the cells were seeded (1×106 cells/well ), exposed with ZnO NPs and incubated for 24h. 15 μL/ well of MTT solution was added to the cells for 2 h. For stop the reaction 100μl of DMSO was added to each well and absorbance was recorded at 570 nm.The decrease of ABTS• absorption is revealing the potent of the ZnO NPs to scavenge ABTS free radicals. Concentration of 500 μg/ml presented maximum ABTS radical scavenging capacity (59.6%) which is significantly lower than the BHA as an standard. In current investigation, ZnO NPs with IC50 about 60 μg/ml inhibitrd ABTS free radicals. Also, It was found that the ZnO NPs suppressed the viability of HT-29 cells (84%) when it was exposed with high concentration (500 μg/ml). IC50 was reported about 200 μg/ml. In the current survey , the obtained results from in vitro trials, confirmed that ZnO NPs of Origanum majorana have a noteworthy effect on antioxidant and antiproliferative properties. Therefore, it may be possible to use such NPs to control and prevent the progression of cancer.
