With the increasing development of nanotechnology and the synthesis of nano materials with specific biological and drug properties, much attention has been drawn to the toxicity of nanoparticles and theireffects on inhibition of cancerous cells. Cancer is a lethal disease that occurs due to the collapse of the balance between cell proliferation and cell death. One of the most important strategies for cancertreatment is the induction of apoptosis. Therefore, identifying different strategies of compounds for suppression of cancer cells is one of the most important research fields. In this survey, the cytotoxicproperties and pro-apoptotic effects of cerium oxide (CeO2) nanoparticles in cultured human brest cancer cells was investigated.Human breast cancer cells (MCF7) were used to evaluate the cellanticancer properties and cytotoxic effects of CeO2-NPs. The MTT method was employed to measure cell viability after exposure to (1.6-50 μg/ml) of CeO2 nanoparticles for 24 h. Acridene orangepropodium iodide (AO/PI) staining was used to evaluate the proapoptotic effect of nanoparticle on Mcf7 cells. For this purpose, the stock solution was made from AO (5mg/ml PBS) and PI(3 mg/ml)stain. To make a work solution 1 μl of each color was added to 1 ml of PBS . Fluorescence microscopy was used to observe the cells. For this purpose, 10 μl of cell suspension was added to the equal volume of dye working solution and observed under the microscope. Cell survival reduced significantly as a function of cerium nanoparticle concentration exposure with IC50 about 50 μg/ml 24h after treatment. Acridine Orange is a vital color that results in the production of green color in living cells but in contrast, PI effects on dead cells and leads to the formation of orange to red color. The results of thefluorescence microscope showed that in untreated cells all cells were green color, while, by treating thecells with CeO2-NPs and increasing the concentration of treatment, the cells with green color wasreduced and some cells are seen in orange and red color which indicates the occurrence of apoptosis and necrosis.The current survey, explores the potential cytotoxic and proapoptotic activity of CeO2-NPs synthesized on MCF-7 cancer cell lines. Considering the biocompatible properties of cerium nanoparticles and the inhibitory effects of the nanoparticle Synthesis by the Rubia tinctorum Plant on cancer cells, it can be used as an adjuvant agent in the treatment and prevention of cancer progression.
