Investigation the toxicity effects of zinc oxide (zno) nanoparticles synthesized in hyssopus officinalis plant extract on the liver cancer and normal cell lines

Cancer is reported as the one of the leading causes of death in the world. Nanotechnology represents a new platform that promises to provide a broad range of novel uses for biological and biomedicalapplications. Metal oxide nanoparticles, including zinc oxide, have advantage of application in biomedicine and as therapeutic agents. In this study, we synthesized Zn O nanoparticles using Hyssopus officinalis plant extract and compared their toxicity on liver cancer and normal cell lines. The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) cell viability assay was done on liver cancer cell line and HUVEC cells as normal cell line. Cells were seeded on 96-well plates and incubated for 24 hours in 37 ºC with 5% humidity. After 24 hours cells were treated with different concentrations of Zn O nanoparticles and incubated for next 24 hours. Then, treatments removed and MTT was added to each well. DMSO was used in order to dissolve insoluble formazan dye and absorbance was read at 570 nm. The inhibitory effects of treatment on cancer cell line and normal cell line were compared. Results showed that Zn O nanoparticles are more potent in killing cancer cells in comparison with normal cell. The values of IC50 for liver cancer cell line and HUVEC cell line were 40 and 70 μg/ml, respectively. Results of this study indicated selective inhibitory effects of Zn O nanoparticles synthesized in hyssop plant extract on cancer cells over normal cell line. These results demonstrate the anticancer potential of Zn O nanoparticles, which could be used as therapeutic agent in cancer therapy
